Sickle Cell Anemia

Sickle cell anemia is an autosomal recessive hereditary disease. The amino acid glutamic acid at position 6 of the β-peptide chain of the patient's hemoglobin is replaced by valine to form sickle-shaped red blood cells and replace the normal hemoglobin. The common clinical manifestations of sickle cell anemia include anemia, jaundice, hepatosplenomegaly, and infection. Since sickle cell anemia is an inherited disease, there is currently no specific cure method, mainly through blood transfusion, drug therapy, and bone marrow transplantation. The prognosis of homozygous patients is poor, and the prognosis of heterozygous patients is relatively good.  

Diagnosis of Sickle Cell Anemia

The basic diagnosis methods of sickle anemia are as follows:

• In vitro sodium bisulfite sickle test. Add 2% sodium bisulfite or 1% sodium nitrite to the blood to make a wet sheet. A large number of sickle-shaped red blood cells can be seen on the cover sheet, which is helpful for diagnosis.
• Blood test. Anemia can be found through blood routine tests.
• Genetic diagnosis. By PCR/restriction enzyme analysis, the 294bp β globin gene amplified fragment cannot be chopped up by OxaNI enzyme. Only the 294bp fragment can be seen in the electropherogram, while the amplified fragment of normal people can be cut into two fragments of 191bp and 103bp by this enzyme. Southern blot hybridization analysis can also be performed by synthesizing specific oligonucleotide probes.
• MRI. MRI signals can effectively observe the involvement range and severity of bone marrow tissues and lesions.
