Human Metapneumovirus Infection

Human metapneumovirus (HMPV) is a new human respiratory virus discovered by Dutch researchers in children with unexplained respiratory infections in 2001. HMPV is a member of the Paramyxoviridae virus and has an RNA genome encoding eight viral proteins. The incidence is higher in infants, the elderly, and those with immune deficiency. The clinical symptoms are similar to those of respiratory syncytial virus infection. Common symptoms after HMPV infection include runny nose, cough, fever, dyspnea, which can cause rhinitis, laryngitis and other upper respiratory tract symptoms, bronchiolitis, pneumonia and other lower respiratory tract symptoms, as well as meningitis, otitis media, etc. Vaccines are the key to preventing HMPV infection.

Diagnosis of Human Metapneumovirus Infection

The experimental diagnosis methods of HMPV include virus isolation, serological diagnosis, RT-PCR (reverse transcription-polymerase chain reaction) and enzyme-linked immunological amplification hybridization analysis, etc. HMPV does not proliferate in most of the cells cultured with respiratory viruses, and it grows rather slowly even in suitable host cells. Therefore, it is difficult to isolate live HMPV virus by cell culture method. The traditional immunofluorescence detection method is simple and fast, but it is limited to the detection of HMPV-specific IgG, which has lower specificity than RT-PCR. RT-PCR technology can amplify and sequence the nucleocapsid protein gene, matrix gene, fusion gene, and polymerase gene of the HMPV virus, and it is currently the main diagnostic method. Compared with traditional detection methods, RT-PCR detection methods have higher diagnostic sensitivity.
