Bocavirus was first discovered in animals in the early 1960s. In August 2005, Swedish scientists used molecular virus screening methods to find a new type of human parvovirus in children’s respiratory secretions for the first time, named human bocavirus (HBoV). HBoV is highly contagious, with high incidence in autumn and winter. HBoV is closely related to human acute respiratory tract infection and gastrointestinal infection, and may cause high fever, dry cough, dyspnea, asthma, acute gastroenteritis and other symptoms.
Diagnosis of Human Bocavirus Infection
HBoV pathogen detection methods are mainly electron microscope, virus isolation and culture method, molecular biology and immunology methods:
- Electron microscopy. The auxiliary means of HBoV detection can observe virus-like particles and guide virus structure and pathogenicity analysis.
- Virus isolation and culture. This method can simulate the natural growth process of HBoV, detect a variety of different viruses while observing the cytopathic effect, and is expected to discover unknown viruses, obtain virus strains, and study their physiological characteristics as well as pathogenic mechanisms.
- Molecular biology detection. Including PCR and real-time PCR detection methods. PCR detection has the advantages of rapidness and sensitivity, and can quickly detect pathogens in samples. Therefore, it has become the most commonly used method for clinical specimen screening for HBoV. TaqMan probe real-time PCR has the characteristics of open technology, strong specificity, and can be used for multiple detections, so it has the most extensive application.
- Immunological test. HBoV infection is dominated by IgM antibodies in the early stage, but disappears more quickly, and then replaced by IgG antibodies with higher yield and longer duration. Thus, the type of antibody (IgM or IgG) and the change in antibody titer can be used to determine whether there is an HBoV infection. Methods of detecting antibodies generally include indirect enzyme-linked immunosorbent assay, western blotting and immunofluorescence detection.
