Chemiluminescence Immunoassay

Chemiluminescence immunoassay (CLIA) was born in 1977. Halman et al. developed a method that combined a highly sensitive chemiluminescence assay technique with a highly specific immune response. CLIA technology labels luminescent substances or enzymes on the antigens or antibodies. After the immune reaction, oxidants or enzyme substrates are added to emit light. The intensity of the emitted light is measured and the concentration of the test substance is determined according to the standard curve. CLIA is a detection and analysis technology for various antigens, haptens, antibodies, hormones, enzymes, fatty acids, vitamins and drugs. It is a new immunoassay technology developed after RIA, IFA and ELISA.

The Workflow of CLIA

CLIA contains direct chemiluminescence immunoassay, chemiluminescence enzyme immunoassay and electrochemiluminescence immunoassay according to different luminescence mechanisms. First, horseradish peroxidase (HRP) and other enzymes are used to label the antigen or antibody. After the immune response, luminol, which is a luminescent substrate, emits light through a redox reaction under the immune action of the enzyme label immune-reactant and the luminescent reagent. Then determine the antigen or antibody by the luminescence intensity.

Applications of CLIA

CLIA uses a microplate photometer to provide a sensitive, high-throughput, and economical method to quantitatively measure antigens in cell plasma, lysates, saliva, tissue, urine, and culture media samples. CLIA has a wide range of applications. It can detect antigens, haptens and antibodies of different molecular sizes, as well as nucleic acid probes. CLIA can be used for clinical diagnosis, environmental monitoring and drug analysis due to its safety, high sensitivity and good specificity.
