Trefoil factor 2 (TFF2) is a small secreted protein. It contains two trefoil domains and has a compact structure. This may be the reason for its high resistance to the harsh environment of the gastrointestinal tract. TFFs are most abundantly expressed in the gastrointestinal tract (especially stomach, colon and pancreas), and they co-localize with mucin. TFF2 is usually co-localized with MUC6 and may be the same as other members of the TFF family, mediating mucin cross-linking and stabilization of the mucin layer. A study of patients with Crohn's disease and inflammatory bowel disease showed that the level of TFF2 in the serum is elevated in an inflammatory state. Another study found that TFF2 levels are higher in patients with sepsis and are associated with the prognosis of sepsis. TFF2 is highly expressed in skeletal muscle, liver, heart, spleen, thymus, lymph nodes and bone marrow. The exact function of TFF2 has not been revealed, but there is evidence that it is related to modulated immune response (TFF2-deficient mice show significant changes in the expression of multiple genes involved in my class of MHC antigens), allergy and asthma.
