This gene (v raf 1 murine leukemia viral oncogene homolog 1) is a cellular homologous gene of the viral raf gene (v-raf). The encoded protein is MAP3K, which acts downstream of the membrane-associated GTPases of the Ras family and binds directly to it. Once activated, the cellular RAF1 protein can phosphorylate and activate the dual-specific protein kinases MEK1 and MEK2, which in turn phosphorylate activates the serine/threonine-specific protein kinases ERK1 and ERK2. Activated ERKs are pleiotropic factors of cell physiology and play an important role in the regulation of cell division cycle, apoptosis, cell differentiation and cell migration and other related gene expression regulation. Mutations in this gene are related to Noonan syndrome 5 and Leopard syndrome 2.
