PML has a tumor suppressor effect. It plays a role in transcriptional regulation, DNA damage response, DNA repair and chromatin organization. PML is involved in the process of retinoic acid regulation, the regulation of cell division, the terminal differentiation of myeloid precursor cells and the differentiation of neural precursor cells. It regulates the regionalization of PTEN by inhibiting usp7-mediated deubiquitination. The PML gene synthesizes a large number of alternative splicing transcripts and produces a variety of PML proteins with a molecular weight of 48-97 kDa.
