FANCD2, also known as FACD and FACD2, is necessary to maintain chromosome stability. FANCD2 participates in the repair of DNA double-strand breaks through homologous recombination and single-strand annealing. After DNA damage, it may participate in the activation of S and G2 checkpoints. It promotes the loading of BRCA2/FANCD1 onto damaged chromatin. FANCD2 may also participate in b-cell immunoglobulin isotype conversion. The defect of FANCD2 is the cause of Fanconi anemia (FA), which is a hereditary heterogeneous autosomal recessive genetic disease characterized by progressive pancytopenia, a variety of congenital malformations and susceptibility to malignant tumors.
