Programmed cell death 1 (PD-1, also known as CD279, Lupus La protein) is an immunosuppressive receptor that belongs to the CD28/CTLA-4 subfamily of the Ig superfamily. It is a type I transmembrane protein of 288 amino acids (aa). PD-1 is expressed during thymus development, and is induced in various surrounding hematopoietic cells through antigen receptor signaling and cytokines. The combination of PD-1 and its ligand PD-L1 or PD-L2 can transduce signals that inhibit T cell proliferation, cytokine production and cytolytic function. It is essential for the regulation of T cell function during immunity and tolerance. PD-1 blockade can overcome immune resistance and has been shown to have anti-tumor activity.
