CDNK1A (Melanoma differentiation-associated protein) may be involved in the inhibition of cell proliferation by p53/TP53-mediated DNA damage. Binding and inhibiting the activity of cyclin-dependent kinases, preventing the phosphorylation of key cyclin-dependent kinase substrates and blocking cell cycle progression. Participate in the nuclear localization and assembly of the cyclin D-CDK4 complex, and promote its kinase activity on RB1. At a higher stoichiometric ratio, the kinase activity of the cyclin D-CDK4 complex is inhibited. It inhibits the synthesis of DNA polymerase delta by competing with POLD3 for PCNA binding.
