CDKN1A (P21) is a cyclin-dependent kinase inhibitor, which is necessary for proper control of the cell cycle and premature aging. P21 is a cyclin-dependent kinase (CDK) inhibitor that inhibits proliferation by inhibiting the activities of CDK2 and CDK1 during the transition of G1/S and G2/M. P21 is the main mediator that p53 induces cell cycle arrest in G1. P21 can interact with proliferating cell nuclear antigen (PCNA) and play a regulatory role in S-phase DNA replication and DNA damage repair. According to reports, P21 is specifically cleaved by casp3-like caspases, leading to significant activation of CDK2, and may contribute to cell apoptosis after caspase activation.
