The function of NCK family adaptor proteins is to connect tyrosine phosphorylation signals to regulate the reorganization of the actin cytoskeleton, leading to cell movement. The cytoplasmic protein NCK1 is an adaptor protein associated with tyrosine phosphorylated growth factor receptors or their cellular substrates. NCK1 binds to many intracellular proteins and affects a variety of signaling pathways, including the gtpase activator protein of Ras, which is responsible for the down-regulation of Ras. NCK1 can regulate UPR, and UPR can be secondary to obesity, impairing glucose homeostasis and the effects of INS.
