Application: | Thrombopoietin (TPO) is produced in the liver by both parenchymal cells and sinusoidal endothelial cells, as well as in the kidney by proximal convoluted tubule cells. It is a primary regulator of megakaryocyte development and platelet production in mammals. TPO binds the TPO receptor, activates JAK and STAT pathways, thus stimulating megakaryocyte growth and platelet production. Additionally, as TPO is vital for the maintenance of haematopoietic stem cells, it can truly be described as a pan- haematopoietic cytokine. |
This test kit is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The TNFSF11 ELISA kit is to be used to detect and quantify protein levels of endogenous TNFSF11. The assay recognizes mouse TNFSF11. An antibody specific for TNFSF11 has been pre-coated onto the microwells. The TNFSF11 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for TNFSF11 is added to detect the captured TNFSF11 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450nm with the correction wavelength set at 630 nm .
