MCL1 is an anti-apoptotic member of the Bcl-2 family and was originally isolated from the ML1 human bone marrow leukemia cell line. Similar to BCL2 and BCL2L1, MCL1 can interact with BAX and/or BAK1 to inhibit mitochondrial-mediated apoptosis. MCL1 protein is essential for the proliferation and survival of myeloma cells in vitro, and the overexpression of MCL1 protein in myeloma cells is related to the recurrence and short event-free survival of patients with multiple myeloma. Recent studies have shown that MCL1 is up-regulated in many hematological and solid tumor malignancies. Therefore, MCL1 is considered as a potential new therapeutic target.
