LARG, also known as ARHGEF12, is one of the three regulators of RhoGEFs in the G protein signal transduction (RGS) domain. LARG activates the rock pathway downstream of the Gα12/13 signal, leading to cytoskeleton reorganization. LARG exerts this activity in the form of homodimer or heterodimer with Arhgef11. LARG is associated with many diseases, including cancer, heart disease and HIV replication. LARG is phosphorylated and dephosphorylated during mitosis, and Cdk1 is the mitotic kinase responsible for the phosphorylation of LARG.
