HOPX (Homeodomain-only protein) gene has many synonyms, including HOD, HOP, OB1, LAGY and NECC1. The protein encoded by this gene is an unusual homeodomain protein that lacks certain conserved residues required for DNA binding. HOPX has different effects on heart growth. The role of Hopx in mouse models is related to myocardial hypertrophy, dilatation and fibrosis. HOPX protein acts as an antagonist of serum response factor (SRF), regulating the opposite process of cell proliferation and differentiation. Overexpression of HOPX leads to cardiac hypertrophy. HOPX protein can inhibit srf-dependent transcriptional activation by recruiting histone deacetylase (HDAC) activity.
