HER2, also known as ERBB2 and Neu, is a 185-kDa transmembrane glycoprotein and a member of the receptor tyrosine kinase epidermal growth factor (EGF) receptor family. It does not have a ligand binding domain by itself, so it cannot bind to growth factors. However, it does bind tightly with other ligand-bound EGF receptor family members to form heterodimers, stabilize ligand binding, and enhance kinase-mediated activation of downstream signaling pathways, such as those involving mitogen activated protein. Kinase and phosphatidylinositol-3 kinase signaling pathway. HER2 amplification and/or overexpression have been reported in many diseases including breast cancer, leukemia and ovarian cancer.
