TSPYL2 is also known as CDA1, and is a new member of the nucleosome assembly protein superfamily. TSPYL2 binds to histones and promotes nucleosome assembly. TSPYL2 is expressed in various tissues, highly expressed in the pituitary gland, and moderately expressed in the adrenal gland, brain, testis and ovary. Immunohistochemical staining analysis of TSPYL2 showed that the cytoplasmic and nuclear staining of several cell types were different. Down-regulation of TSPYL2 expression was found in multiple tumors, suggesting that it has a tumor suppressor effect. Although the molecular weight of TSPYL2 is predicted to be 79.43 kDa, western blot analysis found that the molecular weight of mammalian TSPYL2 is 120 kDa. The abundant acidic amino acid region in TSPYL2 may be the reason for its abnormal migration. In addition, the TSPYL2 protein is unstable and sensitive to proteasome degradation.
