Genetic mutations in the BRCA1 and BRCA2 genes increase the lifetime risk of breast or ovarian cancer. Both BRCA1 and BRCA2 are involved in maintaining genomic stability, particularly homologous recombination pathways for double-stranded DNA repair. BRCA2 proteins contain multiple copies of the 70 AA motif known as the BRC motif, which mediate binding to the RAD51 recombinase, which is involved in DNA repair. BRCA2 is considered a tumor suppressor gene because tumors with BRCA2 mutations often present with wild-type allelic heterozygosity loss.
