ATR is a serine/threonine protein kinase that activates checkpoint signals during genotoxic stress such as ionizing radiation (IR), ultraviolet (UV) or DNA replication stagnation, thereby acting as a DNA damage sensor. ATR and TP53/ p53, which jointly inhibits DNA replication and mitosis, promotes DNA repair, recombination and apoptosis. ATR phosphorylates Ser-139 of the histone variant H2AX/H2AFX at the site of DNA damage, thereby regulating the DNA damage response mechanism. It is essential to maintain the stability of fragile sites and effectively regulate centrosome replication. ATR deficiency is one of the causes of Seckel syndrome type 1 (SCKL1).
