Amphiphysin (AMPH) is a synaptic vesicle-associated protein highly concentrated in nerve endings, and is an autoantigen for breast cancer tough guy syndrome. AMPH contains an n-terminal BAR domain and an endocytic domain, and a c-terminal SH3 domain. In the endocytosis of synaptic vesicles, AMPH is considered to be the linker between clathrin coat and dynamic protein. It may also be involved in the regulation of synapses and the extracellular secretion mechanisms of certain endocrine cell types.
