MLLT3, also known as AF9, locates active transcription start sites (TSSs) through the Yeats domain. TSSs recognize active histone markers such as H3K9 acetylation and croponylation. The deleted MLLT3 with the YEATS domain deleted forms a leukemia fusion protein with the N-terminus of MLL1, misleading the MLLT3 interaction complex to induce abnormal gene transcription. MLLT3 also regulates erythroid or megakaryocyte progenitor cells and has been identified as an HSC hub gene during mouse development. Researchers found that the expression of a gene called MLLT3 is closely related to the self-renewal potential of blood stem cells, and the protein produced by the MLLT3 gene provides blood stem cells with the necessary instructions to maintain their self-renewal ability. It works with other regulatory proteins to keep important parts of the blood stem cell mechanism running during cell division.
